At the end of the course, the participants will be able to design experiments using STED microscopy, prepare high-quality microscopy samples and acquire state-of-the-art nanoscopy images. In a point-scanning implementation of STED nanoscopy, more than 10-fold brighter images have been recorded under typical immunofluorescence sample conditions. DyMIN is a concept which facilitates the highest final resolutions at robust, strong signal for biological STED nanoscopy, as shown here by record axial resolutions (∼60 nm FWHM) for the single-lens 3D STED approach ( Fig. 5 ). 2019-08-27 · STED nanoscopy. STED nanoscopy was performed using a quad scanning STED microscope (Abberior Instruments, Göttingen, Germany) equipped with a UPlanSApo 100x/1,40 Oil objective (Olympus, Tokyo Stimulated emission depletion (STED) nanoscopy is one of the most promising super‐resolution imaging techniques for microstructure imaging. Commercial CdSe@ZnS quantum dots are used as STED probes and ≈50 nm lateral resolution is obtained.

To avoid re-excitation, the STED wavelength has to be tuned at the red tail of the emission spectrum of fluorescent probes, leading to high depletion lase … The well-known saying of “Seeing is believing” became even more apt in biology when stimulated emission depletion (STED) nanoscopy was introduced in 1994 by the Nobel laureate S. Hell and coworkers. We presently stand at a juncture. A widespread implementation of laser-scanning stimulated emission depletion (STED) nanoscopy (3 ⇓⇓ – 6) superposes the excitation focal spot with a doughnut-shaped spot of STED light. Abstract We show that far-field fluorescence nanoscopy by stimulated emission depletion (STED) can be realized with compact off-the-shelf laser diodes, such as those used in laser pointers and DVDs. A spatial resolution of 40-50 nm is attained by pulsing a 660 nm DVD-diode. Stimulated emission depletion (STED) nanoscopy plays a key role in achieving sub-50 nm high spatial resolution for subcellular live-cell imaging.
However STED nanoscopy allows us to get closer to visualize these than ever before, revealing structures inside cells not previously visible. As already mentioned in Sect. 1.2.1, STED nanoscopy is based on the idea of limiting the ability of molecules to fluoresce in the immediate vicinity of the geometric focus.

The 'on' state is the first electronically excited fluorescent (singlet) state of a fluorophore whereas the 'off' state is the ground state. We demonstrate live-cell STED microscopy of two protein species using photochromic green fluorescent proteins as markers. The reversible photoswitching of two markers is implemented so that they can be discerned with a single excitation and STED wavelength and a single detection channel. Dual-label STED microscopy is shown in living mammalian cells.
Due to the non-linear dependence of the stimulated emission rate on the intensity of the STED beam, all the fluorophores around the focal excitation spot will be in their off state (the ground state of the fluorophores).
